There are 15 unlockable characters per version, of which 4 are exclusive per version, all of which are from other indie games:
- Gish (X360) - Sticks to Walls, now has his own section of levels in the X360 mode "Teh Internets".
- Tim from Braid (X360) - Can rewind time three seconds
- Commander Video (Bit.Trip series) - Can hover in mid-air, and while the game says that he is from Bit.Trip Runner, he also stars in Bit.Trip Fate.
- Alien Hominid (X360) - Can shoot downwards to slow descent
- Spelunky (X360) - Has an explosive jump
- Flywrench - Can triple jump, and regains jumps when touching walls, but can't wall-jump [1]
- Ogmo from Jumper - Can double jump
- Ninja from N (X360)
- Castle Crashers (X360)
- The Kid I Wanna Be The Guy - Can double jump; his Warp Zone is considered to be the hardest
- Mighty Jill Off - Can airbreak[2]
- Captain Viridian from VVVVVV (PC) - Slow, but can invert his gravity, negating larger jumps.
- Mr. Minecraft from Minecraft (AKA Steve) (PC) - Game Breaker who can make and destroy blocks at will.
- Goo Ball from World of Goo (Non-Steam PC) - acts exactly like Gish, but according to the blog is "cuter"
- Headcrab from Half-Life (Steam only) - acts exactly like Gish, but with more horizontal jump power, only unlockable character who is from a non-Indie game.
- RunMan from Runman: Race Around The World (PC)
- Naija from Aquaria (PC)

- Only the PC version will have a level editor, due to legal, time and money issues on consoles.
- One of the main reasons to make Super Meat Boy instead of any other project was that the original Meat Boy was by far the most popular game by Edmund on Newgrounds.
- Inspirations include: Mortal Kombat, Super Mario World, Street Fighter 2 (arcade, oh yes), and even Ren and Stimpy[3]
